A Conjunctive Effect of Iron and Selenium Status on Glutathiene Peroxidase Activity and Lipid Peroxidation in Rats

Abstract
The combined effects of iron and selenium status on glutathione peroxidase (GSHPx) activity, cytochrome P-450 activity, and lipid peroxidation in the liver and intestinal mucosa of rats were investigated. Six experimental groups (+Se-Fe, -Se-Fe, +Se+Fe, -Se+Fe, +Se++Fe, -Se++Fe) were manipulated for S weeks with 2% dietary iron (++Fe), normal diet (+Fe), or iron-omitted diet (-Fe) and deionized water (-Se) or selenium-supplemented deionized water (+Se).
Body weight of rats was significantly low in iron-overloaded (++Fe) groups (p$lt;0.01), regardless of Se status. There was no significant difference in hemoglobin content among experimental groups. Serum iron and total tissue iron in the small intestine, intestinal mucosa, and liver were dependent on the iron-load (++Fe$gt;+Fe$gt;-Fe). In the liver of iron-overloaded (++Fe) rats, the total iron was significantly lower in selenium-supplemented group than selenium-deficient group (p$lt;0.05). In the liver and intestine, GSHPg activity was significantly higher in all selenium-supplemented groups, compared to selenium-deficient groups (p$lt;0.01). Lipid peroxidation was significantly dependent on iron-load to rats (++Fe$gt;+Fe$gt;-Fe). In iron-deficient and iron-overloaded rats, GSHPx activity in the liver and intestinal mucosa was positively related to the inhibition of lipid peroxidation. There was no significant difference in cytochrome P-450 activity in the liver of rats between experimental groups.
These results indicate that iron-overload enhances lipid peroxidation in tissues by increasing the tissue iron content and that selenium-dependent GSHPx can be protective against the oxidative damage to tissues.
